Yeast model offers clues to possible drug targets for Lou Gehrig's disease

Amyotrophic lateral sclerosis, also called Lou Gehrig's disease, is a devastatingly cruel neurodegenerative disorder that robs sufferers of the ability to move, speak and, finally, breathe. Now researchers at the Stanford University School of Medicine and San Francisco's Gladstone Institutes have used baker's yeast ? a tiny, one-celled organism ? to identify a chink in the armor of the currently incurable disease that may eventually lead to new therapies for human patients.

"Even though yeast and humans are separated by a billion years of evolution, we were able to use the power of yeast genetics to identify an unexpected potential drug target for ALS," said Aaron Gitler, PhD, an associate professor of genetics at Stanford. "Many neurodegenerative disorders such as ALS, Parkinson's and Alzheimer's exhibit protein clumping or misfolding within the neurons that is thought to either cause or contribute to the conditions. We are trying to figure out why these proteins aggregate in neurons in the brain and spinal cord, and what happens when they do."

In 2008, Gitler received a New Innovator award from the National Institutes of Health to use yeast as a model for understanding human neurodegenerative diseases and as a way to identify new targets for drug development.

Gitler is the co-senior author of the research, which will be published online Oct. 28 in Nature Genetics. Robert Farese, Jr., MD, a senior investigator at the Gladstone Institutes, is the other co-senior author. Stanford graduate student Maria Armakola shares co-first authorship with Matthew Higgins, PhD, a postdoctoral scholar at Gladstone.

Most cases of ALS have no clear-cut cause. However, it has recently been shown that an RNA-binding protein called TDP-43 accumulates in clumps in the cytoplasm of spinal cord neurons in many people with the condition, and mutations in this protein have been found in some people with the ALS. Researchers like Gitler and Farese have been able to mimic the disease in yeast by expressing TDP-43 at higher-than-normal levels, which causes the protein to form lethal clumps in the cells' cytoplasm.

"In humans, the progression of the disease can take years before symptoms arise," said Gitler. "But in yeast, we see protein clumping in the cytoplasm within two days and the cells rapidly begin to die." With their model system in place, Gitler and Farese set out to see whether it was possible to protect yeast cells from this effect by tinkering with the function of other proteins in the cell.

In this study, the researchers discovered that blocking the production of a protein called Dbr1 in a yeast model stops the TDP-43 clumping and allows the cells to live normally. The researchers confirmed the results in human nerve cells grown in the laboratory and in rat neurons overexpressing TDP-43.

"In this study we made no assumptions as to how TDP-43 injures cells," said Farese, "but instead screened the whole yeast genome to find genes that might prevent the toxicity. Independently, both our lab and the Gitler lab found that loss of Dbr1, an enzyme involved in RNA processing, could do this."

Dbr1 serves as part of the cellular clean-up crew that mops up the bits of unwanted RNA generated as part of the protein production line. In our DNA, most genes consist of coding regions, called exons, broken up into several segments by non-coding regions, called introns. Cells can make many different, related proteins from the same stretch of DNA by mixing and matching different exons in a process called splicing.

When the DNA is first copied, or transcribed, into RNA, the introns as well as the exons are included. But the cell quickly splices out the introns, which are released into the cytoplasm as little loops, or lariats. Dbr1, in turn, clips the loops to open them and make them accessible to the cell's disposal system.

Blocking the production of Dbr1 causes the RNA lariats to build up in the cytoplasm. The researchers showed ? by creating lariats with a binding site for a fluorescent tracking protein ? that the mutant TDP-43 binds to these excess lariats rather than clumping. The effect is like using a paper towel to mop up a spill on your computer keyboard: binding to the lariats appears to keep TDP-43 from causing havoc elsewhere.

"Normally, TDP-43 is found in the nucleus," said first author Armakola. "But in the diseased cells, it aggregates in the cytoplasm and forms clumps. We developed a novel way to track where these lariats go in living cells, and we saw that when Dbr1 is missing, the lariats act as a sink to sequester TDP-43."

The researchers note that it's still not entirely clear whether the cells die because the mutant TDP-43 is drawing essential RNA transcripts or regulatory molecules away from the nucleus and into the cytoplasm, or because it's not performing its normal RNA-binding function in the nucleus. Both could contribute to the progression of the disease.

The results in the yeast, rodent and human cells, however, suggest that therapeutic approaches aimed at blocking Dbr1 function, or even creating artificial lariat-like formations to draw away the mutant molecule, should be explored.

"Next, we'd like to explore blocking Dbr1 function in animals such as flies, worms and rodents," said Armakola. "We're also interested in identifying small molecule inhibitors of Dbr1."

Whether you agree monogamy is natural or unnatural, for men or women, I think we can all agree monogamy is a choice and so is infidelity. However, Dr. Anderson?s theory is that if you choose to cheat sexually, but not emotionally, it should not be the sole justification to terminate an otherwise happy and?successful?relationship. Since most men cannot have the best of both worlds, they choose the selfish route ? cheating physically, while remaining faithful emotionally. Observing this phenomenon, Dr. Anderson sought to understand why habitually unfaithful men would even want monogamous relationships in the first place. A quote from the article:

Cheating, however, serves men pretty well. An undiscovered affair allows them to keep their relationship and emotional intimacy, and even if they?re busted it?s a lot easier than admitting that they wanted to screw someone else in the first place.

The good doctor alleges that men, even unfaithful men, desire emotional commitment. It is the physical commitment they struggle with because men are naturally designed to crave sex with other women. For example, his non-statistical study of 120 undergrads showed enjoyment of monogamous sex dipped precipitously after only two years. The way cheating men see it, it?s either cheat or don?t cheat, but telling their partners they want sex outside the relationship, or telling their partners that they actually cheated, is viewed as a surefire way of achieving relationship termination. When men cheat for recreational sex ? not affairs ? they DO love their partners. If they didn?t, they would break up with them.

John Legend, who is not a doctor, covered this subject years earlier in his song?Number One?where he croons the following romantic lyrics, ?You can?t say I don?t love you, Just because I cheat on you, Cuz you can?t see all I do, To keep you from knowing the things I do, Like erase my phone, And keep it out of town, I keep it strapped up when I sleep around.?

As a man, the only thing shocking about Dr. Anderson?s (and John Legend) theories on monogamy and infidelity is the fact that they are so accurate. They correctly explain how unfaithful men justify their actions but that doesn?t mean the actions of unfaithful men should be defended?or should they?

(AP) ? A magnitude 7.7 earthquake struck off the west coast of Canada, but there were no reports of major damage. Residents in parts of British Columbia were evacuated, but the province appeared to escape the biggest quake in Canada since 1949 largely unscathed.

The U.S. Geological Survey said the powerful temblor hit the Queen Charlotte Islands just after 8 p.m. local time Saturday at a depth of about 3 miles (5 kilometers) and was centered 96 miles (155 kilometers) south of Masset, British Columbia. It was felt across a wide area in British Columbia, both on its Pacific islands and on the mainland.

"It looks like the damage and the risk are at a very low level," said Shirley Bond, British Columbia's minister responsible for emergency management said. "We're certainly grateful."

The Pacific Tsunami Warning Center lifted its tsunami advisory for Hawaii Sunday morning just before 4 a.m. local time, three hours after downgrading from a warning and less than six hours after the waves first hit the islands.

Meanwhile, the National Weather Service canceled tsunami advisories for Canada, Alaska, Washington, Oregon and California.

Tsunami Warning Center officials said wave heights were diminishing in Hawaii, though swimmers and boaters should be careful of strong or unusual currents. The biggest waves ? about 5 feet (1.5 meters)high ? appeared to hit Maui.

There were no immediate reports of damage, though one person died in a fatal crash near a road that was closed because of the threat near Oahu's north shore.

Hawaii Gov. Neil Abercrombie said the state was lucky to avoid more severe surges.

"We're very, very grateful that we can go home tonight counting our blessings," Abercrombie said.

Dennis Sinnott of the Canadian Institute of Ocean Science said a 69-centimeter (27 inch) wave was recorded off Langara Island on the northeast tip of Haida Gwaii, formerly called the Queen Charlotte Islands. The islands are home to about 5,000 people, many of them members of the Haida aboriginal group. Another 55 centimeter (21 inch) wave hit Winter Harbour on the northeast coast of Vancouver Island.

Canada's largest earthquake since 1700 was an 8.1 magnitude quake on August 22, 1949 off the coast of British Columbia, according to the Canadian government's Natural Resources website. It occurred on the Queen Charlotte Fault in what the department called Canada's equivalent of the San Andreas Fault ? the boundary between the Pacific and North American plates that runs underwater along the west coast of the Haida Gwaii.

In 1970 a 7.4 magnitude quake struck south of the Haida Gwaii.

The USGS said the temblor shook the waters around British Columbia and was followed by a 5.8 magnitude aftershock after several minutes. Several other aftershocks were reported.

The quake struck 25 miles (40 kilometers) south of Sandspit, British Columbia, on the Haida Gwaii archipelago. People in coastal areas were advised to move to higher ground.

Urs Thomas, operator of the Golden Spruce hotel in Port Clements said there was no warning before everything began moving inside and outside the hotel. He said it lasted about three minutes.

"It was a pretty good shock," Thomas, 59, said. "I looked at my boat outside. It was rocking. Everything was moving. My truck was moving."

After the initial jolt, Thomas began to check the hotel.

"The fixtures and everything were still swinging," he said. "I had some picture frames coming down."

Lenore Lawrence, a resident of Queen Charlotte City on the Haida Gwaii, said the quake was "definitely scary," adding she wondered if "this could be the big one." She said the shaking lasted more than a minute. While several things fell off her mantle and broke, she said damage in her home was minimal.

Many on the B.C. mainland said the same.

"I was sitting at my desk on my computer and everything just started to move. It was maybe 20 seconds," said Joan Girbav, manager of Pacific Inn in Prince Rupert, British Columbia. "It's very scary. I've lived here all my life and I've never felt that."

Residents rushed out of their homes in Tofino, British Columbia on Vancouver Island when the tsunami sirens sounded, but they were allowed to return about two hours after the quake.

In Hawaii, the tsunami warning spurred residents to stock up on essentials at gas stations and grocery stores and sent tourists in beachside hotels to higher floors in their buildings. Bus service into Waikiki was cut off an hour before the first waves, and police in downtown Honolulu shut down a Halloween block party. In Kauai, three schools used as evacuation centers quickly filled to capacity.

In Alaska, the initial wave or surge was recorded at 4 inches (10 centimeters), much smaller than forecast, said Jeremy Zidek, a spokesman for the Alaska Department of Homeland Security and Emergency Management. The first wave hit Craig about two hours after the earthquake. Surges at other Alaska communities were later recorded at 6 inches (15 centimeters), while others were much smaller.

Durable orders show business investment still weak

(AP) ? U.S. companies remained cautious in September and held back on orders for long-lasting manufactured goods that signal investment plans. Weak business investment has contributed to slower economic growth.

The Commerce Department said Thursday that orders for durable goods jumped 9.9 a percent last month. While it was the biggest gain in nearly three years, most of the September gain was driven by a tremendous spike in aircraft orders, which are very volatile and plummeted in the previous month.

When taking out transportation, orders rose just 2 percent. And demand for core capital goods, such as machinery and equipment, were unchanged in September. Core capital goods are considered a proxy for business investment plans. Those orders rose only slightly in August after steep declines in July and June.

Durable goods are products expected to last at least three years. Orders can fluctuate sharply from month to month.

Demand is sharply lower this year, which has weakened manufacturing and hampered economic growth. High unemployment and low pay have kept consumers from spending. Businesses have held back on investing in machinery and equipment. And slower global growth has dampened demand for U.S. exports.

Economists are concerned that businesses could pull back further if Congress doesn't reach an agreement to avert steep tax hikes and government spending cuts scheduled to take effect in January.

There is some indication that U.S. factory activity could be improving. A closely watched survey from the Institute for Supply Management, said manufacturing grew in September for the first time in four months. It was buoyed by a jump in new orders.

Americans also boosted their spending at retail businesses in September, buying more cars, appliances and furniture. Stronger consumer spending could help offset declining demand from overseas.

Still, the decline in U.S. manufacturing has slowed job growth. Manufacturing employment dropped by 16,000 in September after falling by 22,000 in August.

The weak job market has been a key topic in this year's presidential election, which is down to its final days. Voters will have one final employment report to consider, which comes out four days before Election Day.

The economy is not growing fast enough to generate much hiring. Growth slowed to a tepid annual rate of 1.3 percent in the April-June quarter, down from 2 percent in the previous quarter. Most economists see growth staying at or below 2 percent in the second half of the year. The Commerce Department will issue its first estimate of growth in the July-September quarter on Friday.

Tokyo governor to quit, contest election with new party

TOKYO (Reuters) - Tokyo Governor Shintaro Ishihara, whose failed bid to buy disputed islands ignited a crisis with China, said on Thursday he was resigning to form a party with the apparent aim of becoming a "third force" in an upcoming general election.

Ishihara's return to national politics creates further uncertainty about the election, which must be held by next August but could take place early next year. He could also complicate strained relations with China.

"I've decided to resign as Tokyo governor as of today due to various circumstances," Ishihara, 80, told a news conference. "I'm thinking about returning to parliament by forming a new party with my allies."

Ishihara, a writer turned politician, announced in April a plan to buy from their private Japanese owners disputed East China Sea islets claimed by both Japan and China. That prompted a counter-bid from the central government, which nationalized the islands in September.

By buying the islands, the Japanese government sought to avert a deeper crisis with China but the plan backfired, triggering violent protests in China and calls for a boycott of Japanese goods.

Japanese firms scaled back sales, production and investment in China, which is Japan's top export market.

Analysts said Ishihara's island gambit had already made major political forces harden their positions on China.

"If you look in terms of what he wanted to accomplish last April, he succeeded immensely. He provoked a dispute and shifted discourse to the right," said Jeffrey Kingston of Temple University's Japan campus.

"I don't think he can become premier, but he can become a real irksome presence in the central government, in parliament."

His new party could create a "third force" in the election race, possibly in alliance with the nationalist mayor of Osaka, Toru Hashimoto. Hashimoto, 43, also wants to break onto the national stage.

DIVISIVE FIGURE

Polls show the main opposition Liberal Democratic Party (LDP) well ahead of the ruling Democrats and on course to regain power it lost three years ago after half a century of nearly uninterrupted LDP rule.

Both Ishihara and Hashimoto want to score well enough to become coalition partners, analysts said, should the LDP fall short of a majority. But big egos and generational differences may get in the way.

Ishihara suggested he wanted to rally several smaller parties around the group he planned to launch.

Waseda University professor Tetsuro Kato was skeptical.

"It's not going to be like Ishihara becoming the axis around which all the other secondary parties get together," he said.

Though popular in Tokyo, where he was elected to a fourth term a year ago, Ishihara remains a divisive figure, admired by many for his rare blunt style, but unpalatable for others because of his tendency to offend.

Last year, he was forced to apologize for suggesting that a devastating March 11 earthquake and tsunami were "divine punishment" for the "egoism" of the Japanese people.

He has also advocated changing Japan's pacifist constitution and developing nuclear weapons as a deterrent, citing potential threats from China and North Korea.

Ishihara rose to prominence in his 20s as a novelist and entered politics in the late 1960s. In 1989, he made waves with his book "The Japan That Can Say No", co-authored with Sony chairman Akio Morita and calling for Japan to become more assertive with the United States.

Plan to Expand FishHawk Sports Complex Gets Residents' Approval ...

Under a new proposal to expand the FishHawk Sports Complex, the soccer program will get the fields it desperately needs, protected gopher tortoises will get a new home and Starling residents will get peace and quiet.

Hillsborough County Parks, Recreation and Conservation director Mark Thornton outlined the plan at a meeting at the Palmetto Club Wednesday night where a show of hands indicated residents approved of the concept.

In an agreement hashed out with the help of FishHawk Ranch developer Newland Communities, the parks department plans to add four new soccer fields north of the existing complex on 20 acres currently owned by Newlands Communities and designated for conservation.

In exchange, the property dedicated to the county west of the complex, abutting the Starling subdivision, would become a conservation lot with a dedicated habitat for gopher tortoises.

The county also negotiated a deal with the Hillsborough County School District to upgrade two playing fields behind Randall Middle School to the east of the sports complex and add lighting so they can be used by the community's soccer and lacrosse clubs.

Before proceeding, the plan requires state approval to amend the Development of Regional Impact for FishHawk Ranch.

But, if all goes well, Thornton said construction of the four new fields could begin in 18 months. In the meantime, the parks department will go ahead and add lighting to the fields behind Randall in time for the FishHawk Lacrosse program to use them in January.

"With this plan, we think the construction costs will be lower and it's much more user-friendly," said Thornton. "I'm excited about the plan."

Starling residents were equally thrilled.

Under the county's original proposal, the soccer fields would have backed up to homes in the neighborhood.

"Thank you, thank you, thank you," said Jeff Dunlap, one of those residents whose home backs up to the property.

"This demonstrates that the process really does work," said Starling resident Jim Inguagiato. "Our voices were heard."

The proposal also received a thumbs up from residents whose children play sports at the FishHawk Sports Complex.

"It's a win-win for everybody," said resident Steve Hayes.

"This will give us the additional space we need," said Joe Kurtz, president of the FishHawk Soccer Club, which has 1,600 players. He said the soccer club is already using the fields behind Randall but must rent light towers to light the fields at night.

With the approval of the residents, Thornton said the county will begin working on the DRI amendment for FishHawk Ranch and the preliminary engineering.

The county also will look into adding turn lanes into the sports complex to make it easier for Starling residents to turn left out of their neighborhood.

The $2.8 million expansion project is being funded with impact fees.

Cheerleading needs sports safety rules, docs say

(AP) ? Cheerleading isn't just jumping and waving pompoms ? it has become as athletic and potentially as dangerous as a sport and should be designated one to improve safety, the nation's leading group of pediatricians says.

The number of cheerleaders injured each year has climbed dramatically in the last two decades. Common stunts that pose risks include tossing and flipping cheerleaders in the air and creating human pyramids that reach 15 feet high or more.

In a new policy statement released online Monday in the journal Pediatrics, the American Academy of Pediatrics says school sports associations should designate cheerleading as a sport, and make it subject to safety rules and better supervision. That would include on-site athletic trainers, limits on practice time and better qualified coaches, the academy says.

Just like other athletes, cheerleaders should be required to do conditioning exercises and undergo physical exams before joining the squad, the new policy says.

"Not everyone is fully aware of how cheerleading has evolved over the last couple of decades. It used to be just standing on the sidelines and doing cheers and maybe a few jumps," said Dr. Cynthia LaBella, a sports medicine specialist at Chicago's Lurie Children's Hospital and an author of the new policy.

But she said cheerleading often results in injuries that include severe sprains, broken arms and legs, neck injuries and concussions.

Last year, there were almost 37,000 emergency room visits for cheerleading injuries among girls aged 6 to 22, according to data from the Consumer Product Safety Commission. That's more than four times higher than in 1980, when cheerleading was tamer.

While there are still traditional cheerleading squads that support schools' athletic teams, some schools and private clubs have separate cheerleading teams that compete against other teams.

Kali Wald of Elburn, Ill., suffered a serious concussion last year during an acrobatic routine with her high school's competitive team; teammates tossed her in the air but she landed wrong twice, first on her upper back and neck, then on her head. She blacked out for several minutes.

Her father, Dave Wald, said her coaches didn't realize she was seriously injured and never called an ambulance. She still has short-term memory loss and can't attend school full-time because of dizziness, headaches and other concussion symptoms.

Kali, 18, said she believes that cheerleading should be considered a sport and made safer.

Her father agreed and said there needs to be better awareness about the rigors of cheerleading and the potential risks.

Injuries have increased as cheerleading has become more popular. Data suggest there are more than 3 million cheerleaders nationwide aged 6 and older, mostly girls. That includes about 400,000 in high school, according to data cited in the new policy.

While the overall injury rate in high school cheerleading is lower than in other girls sports, including gymnastics, soccer and field hockey, the rate of catastrophic injuries like skull fractures and paralyzing spine injuries is higher, the academy noted.

Kasey Bronstein, 14, and her sister Kori, 17, of Mahwah, N.J., both tore a knee tendon while cheerleading for a private competitive team run by their parents. They twisted their knees doing acrobatic moves while standing on the raised-up hands of their teammates. They had knee surgery last November, followed by extensive physical therapy, and have returned to cheerleading.

Both said it should be considered a sport but they also think it's already pretty safe.

"They're kind of making it too safe, taking out skills that are very exciting to do," Kori said. That includes a double flip stunt no longer allowed on her team.

Some schools and state high school sports associations already consider cheerleading a sport and require the kind of safety oversight that the academy is recommending. But many do not, said Jim Lord, executive director of the American Association of Cheerleading Coaches & Administrators. Some don't consider it a sport because not all cheerleading squads are involved in their own competitions, he said.

Lord said the academy's policy mirrors many of his group's safety recommendations for high schools and colleges.

That includes limiting the height of human pyramids in high school cheerleading to just two people. The academy also says routines that include pyramids, tumbling or tosses should not be performed on hard surfaces.

Lisa Kluchorosky, a sports medicine specialist who works with the academy and the National Athletic Trainers Association, said the new policy will help erase misconceptions that cheerleading is not very athletic.

"The statistics are compelling and you can't turn your head from that," she said.
